The contract also includes a buyout clause worth €60 million.
Confirmation of the deal comes after club president Josep Maria Bartomeu said last month that Barcelona had agreed to a €25-million fee to secure the French defender's services.
A statement on the team's website read: "The player comes to FC Barcelona to reinforce the defence for Luis Enrique's team. The Frenchman can play at the centre of defence or at left back. Despite his youth, he has played 170 games for Olympique, acting as captain on several occasions."
The club announced that Umtiti will arrive in Barcelona on Thursday for his unveiling, just days after competing at Euro 2016 and playing in the final during France's loss to Portugal on Sunday.
The centre-back made 30 Ligue 1 appearances for a Lyon side that finished second to champions Paris Saint-Germain.
